I would proceed as follows:
.NET has a class called "SmtpClient" that makes sending out email easy. Following is the code structure/process what I roughly got in mind
1) Create a new class that have helper functions like "SendUserVerificationEmail(UserObject)". These functions will prepare email body and subject and will utilize underlying SmtpClient class to send out the actual email. All the configuration required to configure SmtpClient (e.g. Smtp host, port , username, password) will be stored in a config file so they can be modified without recompiling the app/web.
2) Verification email body will have URL/Link that will be dynamically generated everytime the email is sent. And that link will be tied to the User to whom the email is sent.
3) Once user click on the link the system will mark this email address "system_verified" so that the Administrator can know that automated system has verified this email and then they can manually approve the email and it is marked as "verified".

***Identifying non-active email address is not a straightforward process. There are few tricks that we can implement but they are not 100% correct. We can discuss it after starting.***
